PSI-12 Dietary isoacids modulate nutrient digestibility and fermentation products in ileal digesta and feces of growing pigs fed corn-soybean meal diets

Abstract Branched-chain fatty acids, or isoacids, are the product of branched-chain amino acid fermentation and have been associated with benefits on the productivity of ruminants, primarily by positively modulating gut microbes and promoting gastrointestinal health. The objective of this study was to determine the optimal inclusion level of isoacid blend product based on apparent ileal digestibility (AID), apparent total tract digestibility (ATTD) of nutrients, and fermentation products in growing pigs fed corn-soybean meal diets. Twelve cannulated pigs [body weight (BW) = 20.9 + 0.6 kg] were used in a 5-period switch-back design with 6 diets and 2 replicate pigs in each period (n = 10 observations per dietary treatment). Experimental diets consisted of increasing levels (0%, 0.5%, 0.75%, 1%, 1.25%, and 1.5%) of an isoacid blend (isobutyrate, isovalerate, and 2-methyl butyrate, 1:1:1) added to a corn-soybean meal basal diet containing titanium dioxide as an indigestible marker. Each experimental period lasted 14 d: 10 d for acclimatization to the diets, 2 d for fecal collection, and 2 d for digesta collection. Diets, fecal, and ileal digesta samples were analyzed for gross energy (GE), crude protein (CP), fiber composition, amino acids (AA), and titanium. The concentration of short-chain fatty acids and ammonia were analyzed in digesta and fecal samples. Orthogonal polynomial contrasts were used to determine the linear and quadratic effects of increasing levels of isoacids. Dietary supplementation of isoacids improved (P < 0.1, linear and/or quadratic) AID of crude fiber and hemicellulose and most of the indispensable AA (except Met+Cys, Trp, and Val) as well as ATTD of CP, GE, neutral detergent fiber, and hemicellulose. In addition, ATTD of Arg, His, Ile, Leu, Met+Cys, Phe, Thr, Trp, and Val increased quadratically (P < 0.1). Collectively, 1% isoacid inclusion promoted the greatest responses in most measured variables (% increase: unsupplemented vs. 1%): AID of Lys (9.3%), Met (5.7%), Thr (17.8%), Leu (7.4%), Ile (9.2%), Phe (7.8%), His (5.9%), Arg (4.9%); ATTD of CP (2.1%), GE (1.3%), NDF (3.9%), hemicellulose (>50%), overall indispensable AA (9.5%), overall dispensable AA (8.6%). On the other hand, the inclusion of isoacids quadratically increased (P = 0.071) the concentration of ammonia and decreased (P < 0.1) the concentration of acetic acid, iso-valeric acid, and total VFA in ileal digesta samples. In fecal samples, the proportion of acetic acid decreased (P < 0.05) quadratically while the proportion of propionic, valeric, and caproic acids increased (P < 0.1). In conclusion, dietary supplementation of isoacids at 1% can improve dietary nutrient digestibility, particularly ileal AA digestibility. Based on the changes in fermentation products, the modulation of intestinal microbiota through the gastrointestinal tract could be potentially related to the effects of isoacids on nutrient digestibility. However, the effects of isoacids supplementation on intestinal microbiota must be determined.

Effect of in ovo feeding of xylobiose and xylotriose on plasma immunoglobulin, cecal metabolites production, microbial ecology, and metabolic pathways in broiler chickens

BackgroundDietary supplementation of xylooligosaccharides (XOS) has been found to influence gut health by manipulating cecal microbiota and producing microbe-origin metabolites. But no study investigated and compared the effect of in ovo feeding of xylobiose (XOS2) and xylotriose (XOS3) in chickens. This study investigated the effect of in ovo feeding of these XOS compounds on post-hatch gut health parameters in chickens. A total of 144 fertilized chicken eggs were divided into three groups: a) non-injected control (CON), b) XOS2, and c) XOS3. On the 17th embryonic day, the eggs of the XOS2 and XOS3 groups were injected with 3 mg of XOS2 and XOS3 diluted in 0.5 mL of 0.85% normal saline through the amniotic sac. After hatching, the chicks were raised for 21 d. Blood was collected on d 14 to measure plasma immunoglobulin. Cecal digesta were collected for measuring short-chain fatty acids (SCFA) on d 14 and 21, and for microbial ecology and microbial metabolic pathway analyses on d 7 and 21.ResultsThe results were considered significantly different at P < 0.05. ELISA quantified plasma IgA and IgG on d 14 chickens, revealing no differences among the treatments. Gas chromatography results showed no significant differences in the concentrations of cecal SCFAs on d 14 but significant differences on d 21. However, the SCFA concentrations were lower in the XOS3 than in the CON group on d 21. The cecal metagenomics data showed that the abundance of the family Clostridiaceae significantly decreased on d 7, and the abundance of the family Oscillospiraceae increased on d 21 in the XOS2 compared to the CON. There was a reduction in the relative abundance of genus Clostridium sensu stricto 1 in the XOS2 compared to the CON on d 7 and the genus Ruminococcus torques in both XOS2 and XOS3 groups compared to the CON on d 21. The XOS2 and XOS3 groups reduced the genes for chondroitin sulfate degradation I and L-histidine degradation I pathways, which contribute to improved gut health, respectively, in the microbiome on d 7. In contrast, on d 21, the XOS2 and XOS3 groups enriched the thiamin salvage II, L-isoleucine biosynthesis IV, and O-antigen building blocks biosynthesis (E. coli) pathways, which are indicative of improved gut health. Unlike the XOS3 and CON, the microbiome enriched the pathways associated with energy enhancement, including flavin biosynthesis I, sucrose degradation III, and Calvin-Benson-Bassham cycle pathways, in the XOS2 group on d 21.ConclusionIn ovo XOS2 and XOS3 feeding promoted beneficial bacterial growth and reduced harmful bacteria at the family and genus levels. The metagenomic-based microbial metabolic pathway profiling predicted a favorable change in the availability of cecal metabolites in the XOS2 and XOS3 groups. The modulation of microbiota and metabolic pathways suggests that in ovo XOS2 and XOS3 feeding improved gut health during the post-hatch period of broilers.

233 Inclusion of a multispecies fungal feed additive in forage-based diets fed to beef cattle: Effects on growth performance and ruminal fermentation

Abstract The objective was to evaluate the effect of adding a multispecies fungal feed additive (MFA; Aspergillus flavus, Aspergillus niger, Aspergillus oryzae) to forage-based diets fed to beef cattle on growth performance and ruminal fermentation. Experiment 1 evaluated the effect of supplementing Angus × SimAngus-crossbred steers [n = 80; initial body weight (BW) = 370 ± 44 kg] with varying doses of an MFA in a randomized complete block design. Steers were blocked by BW and housed in 8 feedlot pens (randomly assigned 2 pens per treatment) where a hay-based diet was fed for 122 d. The hay-based diet was offered with the inclusion of the MFA at 0.02, 0.04, and 0.08 % of the diet dry matter (DM), or without the inclusion of the MFA (CON). Steers were weighed at 28-d intervals. For experiment 2, ruminally cannulated Angus × SimAngus cows (n = 4; initial BW = 569 ± 21 kg) were used in a crossover design with two 21-d study periods to evaluate the effect of dietary inclusion of an MFA on in situ digestion and rumen metabolites. Treatments consisted of a forage-based diet with (WFA) or without (CON) the inclusion of the MFA. In situ digestion was evaluated on d 5, 10, and 20 throughout each period. In-situ bags were incubated in the rumen for 0 to 36 h, sampling every 4 h. On d 20, samples of ruminal fluid were collected to analyze short-chain fatty acids (SCFA) concentration. All data were analyzed using the PROC MIXED procedure of SAS. For experiment 1, a polynomial contrast was used. For experiment 2, the interaction between period and treatment was not significant (P &amp;gt; 0.45). In Experiment 1, the final BW, average daily gain (ADG), and gain to feed ratio (G:F) changed cubically (P ≤ 0.04; Table 1) as dietary inclusion of the MFA increased. Steers supplemented with the MFA at a rate of 0.02% of the diet DM were heavier at the end of the 122-d study period and had the greatest ADG and G:F among treatments. No differences between treatments (P ≥ 0.37) were observed for dry matter intake. In Experiment 2, no treatment × time interactions and treatment effects were detected (P ≥ 0.17; Table 2) for the in-situ disappearance rate and degradation kinetics of the diet DM, neutral detergent fiber, and acid detergent fiber. In addition, MFA supplementation did not affect (P ≥ 0.13; Table 3) the SCFA profile and the total SCFA content in the rumen fluid. Results from these studies indicate that dietary inclusion of an MFA might be an effective alternative to increase the feed efficiency of beef cattle-fed forage-based diets. However, the precise mode of action warrants further investigations.

168 Effects of feeding barley-based diets differing in fermentable starch and fiber on nutrient digestion and body composition in weaned pigs

Abstract Hulled and hull-less barley grains containing low insoluble fiber are alternative energy sources in weaning diets but are typically not part of weaned pig diets. The range in fermentable starch and fiber content among barley cultivars presents opportunities to investigate whether these fermentable carbohydrates of barley grain do influence nutrient digestion, gut short chain fatty acid (SCFA) profile and body composition. To explore, pigs [n = 40; body weight (BW) = 8.6 kg; 1-wk post-weaning] were housed individually and fed 1 of 4 diets containing 600 to 705 g of cereal grain/kg: 1) low-fermentable wheat (LFW); 2) high β-glucan, hull-less barley (HFBB); 3) high amylose, hull-less barley (HFAB); or 4) low-fermentable hulled barley (LFB). Diets provided 10.2 and 9.8 MJ NE/kg, and 1.32 and 1.21 g standardized ileal digestible (SID) lysine/MJ NE for phase 1 (d 0–14) and phase 2 (d 15–28), respectively. At the end of the trial, feces were collected, and pigs were slaughtered to collect digesta and the carcass that was subsequently ground. Subsamples of feces, digesta, and carcasses were freeze-dried and analyzed. The apparent ileal digestibility (AID) of crude protein (CP) and gross energy (GE) did not differ among diets. The AID of β-glucan was 3 times greater (P &amp;lt; 0.05) for LFW than HFBB and HFAB, while AID of starch was 12% less (P &amp;lt; 0.05) for HFAB than the other diets. Apparent total tract digestibility (ATTD) of GE and CP was 4.2 and 8.9% less (P &amp;lt; 0.05), respectively, for the 3 barley diets than the wheat diet. The apparent hindgut fermentation (AHF) of β-glucan was 2.6 times greater (P &amp;lt; 0.05) for HFAB and HFBB than LFW, while AHF of starch was 7.5 times greater for HFAB than LFW. The calculated NE value was greatest (P &amp;lt; 0.05) for LFW, intermediate for HFBB and HFAB, and least for LFB. Among diets, HFAB increased (P &amp;lt; 0.05) the concentration of butyrate (indicator of gut health) and propionate in cecal and colonic digesta, with butyrate up to 50% more (P &amp;lt; 0.05) than that of LFW. Carcass protein and lipid concentration did not differ among pigs fed the 4 diets. In conclusion, feeding barley instead of wheat grain to weaned pigs reduced diet nutrient digestibility but did not affect carcass protein and lipid concentration. Feeding high amylose barley increased gut SCFA concentration. Barley grain with increasing fermentable carbohydrates can thus be included as alternative ingredient in diets of young pigs.

Fucoidan from Apostichopus japonicus ameliorates alcoholic liver disease by regulating gut-liver axis homeostasis.

Long-term and excessive alcohol consumption can lead to the development of alcoholic liver disease (ALD), characterized by oxidative damage, intestinal barrier injury, and disruption of intestinal microbiota. In this study, we extracted fucoidan (Aj-FUC) from Apostichopus japonicus using enzymatic methods and characterized its structure. The ALD model was established in male Balb/c mice using 56° Baijiu, with silymarin as a positive control. Mice were orally administered 100 mg/kg·bw and 300 mg/kg·bw of Aj-FUC for 28 days to evaluate its effects on liver injury in ALD mice and explore its potential role in modulating the gut-liver axis. The results showed significant improvements in histopathological changes and liver disease in the Aj-FUC group. Aj-FUC treatment significantly increased the levels of glutathione (GSH) and glutathione peroxidase (GSH-Px) while weakly reduced the elevation of malondialdehyde (MDA) induced by ALD. It also regulated the Nrf2/HO-1 signaling pathway, collectively alleviating hepatic oxidative stress. Aj-FUC intervention upregulated the expression of ZO-1 and Occludin, thus contributing to repair the intestinal barrier. Additionally, Aj-FUC increased the content of short-chain fatty acids (SCFAs) and regulated the imbalance in gut microbiota. These results suggested that Aj-FUC alleviates ALD by modulating the gut-liver axis homeostasis. It may prove to be a useful dietary supplement in the treatment of alcoholic liver damage.

Curdlan-polyphenol complexes prepared by pH-driven effectively enhanced their physicochemical stability, antioxidant and prebiotic activities.

In this study, the curdlan-polyphenol complexes were constructed by a pH-driven method. The interaction between curdlan and various hydrophobic polyphenols (curcumin, quercetin, and chlorogenic acid) was investigated. Curdlan could self-assemble into particles for loading polyphenols through hydrogen bonding and hydrophobic interactions. The three polyphenols were embedded in curdlan in an amorphous state. The curdlan-curcumin complex showed the lowest viscoelasticity but exhibited the highest curcumin loading ability (34.04±1.73mg/g). However, the curdlan-chlorogenic acid complex emerged the opposite trend, indicating that the loading capacity was associated with the hydrophobicity of polyphenols. The antioxidant activity of curdlan significantly increased after combining with polyphenols, which could be maintained during in vitro simulated gastrointestinal digestion. In particular, the curdlan-quercetin complex exhibited the highest antioxidant activity and short-chain fatty acid concentration, which could influence gut microbiota composition by promoting the proliferation of Prevotella and inhibiting the growth of Escherichia_Shigella. In conclusion, the curdlan-polyphenol complexes prepared by an alcohol-free pH-driven method could effectively enhance the gastrointestinal stability of polyphenols as well as increase the antioxidant and prebiotic activities of curdlan, which could be applied as a functional ingredient to improve gut health.

Polysaccharides of Aspergillus cristatus attenuate obesity by regulating gut microbiota and gut microbiota-related metabolites

Golden-flower fungus, the only dominant microorganism determining the Fu-brick tea quality through fermentation and the important microbe in Liupao tea, is considered a potential probiotic fungus based on its anti-obesity effect. However, the classification of golden-flower fungi is still controversial; the anti-obesity effect of golden-flower fungus polysaccharides remains unknown. In this study, we identify a golden-flower strain as Aspergillus cristatus based on morphological characteristics and multigene phylogeny analysis, which resolves the controversy of classification. Moreover, we find A. cristatus polysaccharides (ACPS) attenuate obesity in rats. ACPS modulate gut bacterial composition, in which Akkermansia, Akkermansia muciniphila, Bacteroides, Romboutsia, Blautia, and Desulfovibrio are considered the core microbes regulated by ACPS. ACPS increase fecal total short-chain fatty acid content and serum, hepatic, and fecal total bile acid content. Furthermore, ACPS-induced gut microbiota alteration plays a causal role in the protection from obesity, according to a fecal transplantation experiment. Thus, ACPS ameliorate obesity by regulating gut microbiota and gut microbiota-related metabolites.

Effects of Multi-species Synbiotic Supplementation on Circulating miR-27a, miR-33a Levels, and Lipid Parameters in Adult Men with Dyslipidemia; A Randomized, Double-Blind, Placebo-Controlled Clinical Trial.

MicroRNAs (miRNAs) have emerged as important regulators of lipid metabolism. Recent studies have suggested synbiotics may modulate miRNA expression and lipid metabolism. This study aimed to investigate the effects of synbiotic supplementation on circulating miR-27a, miR-33a, and lipid parameters in patients with dyslipidemia. Fifty-six eligible participants were randomly allocated to receive either synbiotic or placebo sachets twice a day for 12 weeks. Each synbiotic sachet contained 3×1010 CFU six species of probiotic microorganisms and 5 grams of inulin and fructooligosaccharide (FOS) as prebiotics. Serum miR-27a and miR-33a expression levels, serum lipids, and apolipoproteins, the fecal concentration of short-chain fatty acids (SCFAs), and Firmicutes and Bacteroidetes phyla were assessed before and after the study. Real-time PCR was used to determine the relative expression levels of miRNAs. The results showed synbiotic supplementation significantly downregulated the expression levels of miR-27a and miR-33a compared to the placebo group (p = 0.008 and p = 0.001, respectively). Furthermore, the intervention group exhibited significant improvements in serum high-density lipoprotein (HDL-C), small dense low-density lipoprotein (sdLDL-C), apoA-I, and apoB-100 (p = 0.008, p = 0.006, p = 0.003, p = 0.001, respectively). The results showed a significant negative correlation between miR-33a expression levels with HDL-C, butyrate, propionate, and a significant positive correlation with total cholesterol (TC), low-density lipoprotein (LDL-C), and sdLDL-C in the intervention group. Fecal bacteria and SCFAs were significantly increased in the intervention group. This study provides evidence that synbiotic supplementation can modulate miR-27a and miR-33a expression and improve lipid metabolism in patients with dyslipidemia.

Effect of Miscellaneous Meals Replacing Soybean Meal in Feed on Growth Performance, Serum Biochemical Parameters, and Microbiota Composition of 25–50 kg Growing Pigs

The present study aims to determine the effect of miscellaneous meals (rapeseed meal, cottonseed meal, and sunflower meal) replacing soybean meal in feed on growth performance, apparent digestibility of nutrients, serum biochemical parameters, serum free amino acid content, microbiota composition and SCFAs content in growing pigs (25–50 kg). A total of 72 (Duroc × Landrace × Yorkshire) growing pigs with initial weights of 25.79 ± 0.23 kg were randomly divided into three treatments. The pigs were fed corn–soybean meal (CON), corn–soybean–miscellaneous meals (CSM), and corn–miscellaneous meals (CMM). Each treatment included six replicates with four pigs per pen (n = 24, 12 barrows and 12 gilts). Soybean meal accounted for 22.10% of the basal diet in the CON group. In the CSM group, miscellaneous meals partially replaced soybean meal with a mixture of 4.50% rapeseed meal, 3.98% cottonseed meal, and 4.50% sunflower meal. In the CMM group, miscellaneous meals entirely replaced soybean meal with a mixture of 8.50% rapeseed meal, 8.62% cottonseed meal, and 8.5% sunflower. The results showed that compared with the CON, the CSM and CMM groups significantly improved the average daily gain (ADG) of growing pigs during the 25–50 kg stage (p &lt; 0.05) but had no effects on average daily feed intake (ADFI) and average daily feed intake/average daily gain (F/G) (p &gt; 0.05). Moreover, the CMM group significantly reduced nutrient apparent digestibility of gross energy compared with the CON group. The serum biochemical parameters results showed that the CSM group significantly improved the contents of total protein (TP) compared with the CON group (p &lt; 0.05). The CMM group significantly improved the contents of total protein (TP), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) compared with the CON group in serum (p &lt; 0.05). In comparison with the CON group, the CMM group also significantly improved lysine (Lys), threonine (Thr), valine (Val), isoleucine (Ile), leucine (Leu), phenylalanine (Phe), arginine (Arg), and citrulline (Cit) levels in serum (p &lt; 0.05). However, the CMM group significantly decreased non-essential amino acid content glycine (Gly) in serum compared with CON (p &lt; 0.05), while compared with the CON group, the CSM and CMM groups had no significant effects on the relative abundance, the alpha-diversity, or the beta-diversity of fecal microbiota. Moreover, compared with the CON group, the CSM group significantly increased butyric acid and valeric acid contents of short-chain fatty acids (SCFAs) in feces (p &lt; 0.05). In contrast to the CON group, the CMM group significantly reduced the contents of SCFAs in feces, including acetic acid, propionic acid, and isobutyric acid (p &lt; 0.05). Collectively, the results of the present study indicate that miscellaneous meals (rapeseed meal, cottonseed meal, and sunflower meal) can partially replace the soybean meal and significantly improve the growth performance of growing pigs during the 25–50 kg stage. Thus, miscellaneous meals are a suitable protein source as basal diets to replace soybean meals for 25–50 kg growing pigs. These results can be helpful to further develop miscellaneous meals as a functional alternative feed ingredient to soybean meal.

Nano-Selenium Alleviates Cd-Induced Chronic Colitis through Intestinal Flora

Background: Cadmium (Cd) is an environmental contaminant that poses risks to human and animal health. Selenium (Se), a beneficial element, alleviates the detrimental consequences of colitis and Cd toxicity. Se is found in food products as both inorganic Se (sodium selenite) and organic Se (typically Se-enriched yeast). Nano-selenium (nano-Se; a novel form of Se produced through the bioreduction of Se species) has recently garnered considerable interest, although its effects against Cd-induced enterotoxicity are poorly understood. The aim of this study was to investigate the impact of nano-selenium on mitigating cadmium toxicity and safeguarding the integrity of the intestinal barrier. Methods: For a total of two cycles, we subjected 6-week-old C57 mice to chronic colitis by exposing them to Cd and nano-selenium for two weeks, followed by DSS water for one week. Results: The application of nano-selenium mitigated the intensity of colitis and alleviated inflammation in the colon. Nano-selenium enhanced the diversity of the intestinal flora, elevated the concentration of short-chain fatty acids (SCFAs) in feces, and improved the integrity of the intestinal barrier. Conclusions: In summary, nano-Se may reduce intestinal inflammation by regulating the growth of intestinal microorganisms and protecting the intestinal barrier.

Whole-plant silage maize to improve fiber digestive characteristics and intestinal microbiota of Hezuo pigs

IntroductionUtilizing roughage resources is an effective approach to alleviate the shortage of corn-soybean feed and reducing the costs in the swine industry. Hezuo pig is one group of plateau type local Tibetan pig with strong tolerance to crude feeding. Nevertheless, current research on the roughage tolerance in Hezuo pigs and the microbiological mechanisms behind it is still minimally.This study explored the impact of various ratios of whole-plant silage (WPS) maize on the pH, cellulase activity, short-chain fatty acids (SCFAs), and intestinal microbiota in Hezuo pigs.MethodsThirty-two Hezuo pigs were randomly divided into four groups (n = 8). The control group received a basal diet, while experimental groups I, II, and III were given diets with incremental additions of 5%, 10%, and 15% air-dried WPS maize, respectively, for 120 days.ResultsThe findings revealed that compared with the control group, in Group II, the pH of cecum and colon were notably decreased (p &amp;lt; 0.05), while acid detergent fiberdigestibility, the concentration of propionic and isobutyric acid in the cecum, and the concentration of isobutyric acid in the colon were significantly increased (p &amp;lt; 0.05). Also, carboxymethyl cellulase activity in the cecum in group II of Hezuo pigs was significantly higher than that in the other three groups (p &amp;lt; 0.05). Furthermore, the cecum microbiota showed a higher diversity in the group II of Hezuo pigs than that in the control group, as shown by the Simpson and Shannon indices. Specifically, 15 and 24 bacterial species showed a significant difference in relative abundance at the family and genus levels, respectively. Correlation analyses revealed significant associations between bacterial genera and SCFAs concentrations in the cecum. The abundance of Bacteroides and NK4A214_group was positively correlated with amounts of valeric and isovaleric acid but negatively with propionic acid (p &amp;lt; 0.05). The abundance of UCG-010 was positively linked with acetic acid and negatively correlated with butyric acid (p &amp;lt; 0.05). Actinobacillus abundance was positively associated with butyric acid levels (p &amp;lt; 0.05).DiscussionIn conclusion, a 10% WPS maize diet improved crude fiber digestibility by lowering cecal and colonic chyme pH, enhancing intestinal cellulase activity, improving SCFA production, and increasing intestinal microbiota diversity.

Performance, digestibility and meat quality from lambs fed diets with plantago lanceolata, Italian ryegrass or sainfoin herbages.

Plantago species, which is known to adapt to different climatic conditions, drought, temperature and different soil types. The aim of this study was to compare the effects of P. lanceolata herbage on fattening performance, rumen variables, digestibility, meat quality, meat nutrients and meat textural variables in lambs with the values of Italian ryegrass and sainfoin herbages. The lambs were fed total mix ration (TMR) with concentrated feed (about 30%) plus forage (about 70%) (P. lanceolata, PLA, Italian ryegrass (Lolium multiflorum); IRY or sainfoin (Onobrychis viciifolia) herbages, SAI). Twenty-four male lambs (Akkaraman breed) were assigned to three treatments with 8 lambs in each group. Fattening performance parameters, digestibility and carcass variables were determined. Meat quality and shelf life variables (oxidation, fatty acid profile and textural variables) was detected in the loin (Musculus longissimus), shoulder (M. deltoideus) and leg (M. semitendinosus) muscle samples, which were rested for 24 h +4°C and stored at -20°C for one week, one month, three months and six months. The dry matter (DM) intake, body weight gain, feed efficiency and carcass yield values of lambs, the DM and organic matter (OM) digestions (DMD and OMD) of TMRs, and the pH value, ammonia-nitrogen and short chain fatty acid (SCFA) concentrations of rumen fluid in lambs consuming PLA were similar to those of IRY and SAI (p > 0.05). PLA increased water holding capacity (WHC) and decreased cooking loss of meat compared to feeding with those of IRY and SAI (p < 0.05). The peroxide value (PV) and malondialdehyde (MDA) concentration increased with the stocking time in the meats (p < 0.05). Therefore, PV and MDA concentrations of shoulder, leg and loin meats up to 6th months for PLA were lower than those of IRY and SAI (p < 0.05). The linoleic acid concentration of loin and shoulder meats in PLA were higher than those of IRY and SAI (p < 0.05). The average oleic acid and ∑ω6 fatty acids concentration of meat in PLA was lower than those in IRY and SAI (p < 0.05). Atherogenic index and thrombogenic index values and palmitic acid levels of meat in PLA were similar to those in IRY, but lower than those in SAI (p < 0.05). Springiness, cohesiveness, resilience, hardness and chewiness values of the textural analysis parameters in meat of PLA were lower than those of IRY and SAI. As a result, effect of P. lanceolata on the fattening performance and rumen fermentation of lambs were similar with those of Italian ryegrass (Gramineae family) and sainfoin (Leguminosae family) forages and, it had a positive effect on meat quality (water holding capacity, cooking loss) and meat shelf life criteria (MDA, PV, fatty acid profile and textural variables). For functional lamb meat production, P. lanceolata can be included in the diet of lamb fattening.

The therapeutic potential of Ziziphi Spinosae Semen and Polygalae Radix in insomnia management: Insights from gut microbiota and serum metabolomics techniques

Ethnopharmacological relevanceZiziphi Spinosae Semen and Polygalae Radix (ZSS-PR) constitute a traditional Chinese herbal combination with notable applications in clinical and experimental settings due to their evident sedative and calming effects. Aligned with traditional Chinese medicine principles, Ziziphi Spinosae Semen supports cardiovascular health, nourishes the liver, and induces mental tranquillity. Simultaneously, Polygalae Radix elicits calming effects, fosters clear thinking, and reinstates proper coordination between the heart and kidneys. ZSS-PR is commonly employed as a therapeutic intervention for various insomnia types, demonstrating distinct clinical efficacy. Our previous study findings provide evidence that ZSS-PR administration significantly reduces sleep onset latency, increases overall sleep duration, and improves abnormal neurotransmitter levels in a murine insomnia model. Aim of studyThis investigation aimed to scrutinize the intrinsic regulatory mechanism of ZSS-PR in managing insomnia using gut microbiota and serum metabolomics techniques. Materials and methodsMice were given DL-4-Chlorophenylalanine to induce insomnia and then treated with ZSS-PR. The open-field test assessed the animals' spontaneous activity. Concentrations of neurotransmitters, endocrine hormones, and cytokines in the duodenum were measured using enzyme linked immunosorbent assay, and brain histopathology was evaluated with H&E staining. The impact of ZSS-PR on the metabolic profile was examined by liquid chromatography couped to high resolution mass spectrometry, and 16S rDNA sequencing was used to study the influence of ZSS-PR on the gut microbiota. Additionally, the content of short-chain fatty acids (SCFAs) was analyzed by GC-MS. Finally, correlation analysis investigated relationships between biochemical markers, metabolites, SCFAs, and gut microbiota. ResultsZSS-PR treatment significantly increased movement time and distance in mice with insomnia and improved pathological impairments in the cerebral cortex and hippocampus. It also restored abnormal levels of biochemical markers in the gut of insomnia-afflicted mice, including 5-hydroxytryptamine, dopamine, gastrin, melatonin, tumour necrosis factor-α, and interleukin-1β. Metabolomics findings showed that ZSS-PR had a significant restorative effect on 15 endogenous metabolites in mice with insomnia. Furthermore, ZSS-PR primarily influenced five metabolic pathways, such as phenylalanine, tyrosine, and tryptophan biosynthesis, glutamine, and glutamate metabolism. Additionally, gut microbiota analysis revealed notable alterations in both diversity and microbial composition after ZSS-PR treatment. These changes were primarily attributed to the relative abundances of microbiota, including Firmicutes, Bacteroidota, Fusobacteriota, Muribaculaceae_unclassified, and Ligilactobacillus. The results of SCFAs analysis demonstrated that ZSS-PR effectively restored abnormal levels of acetic acid, propionic acid, isobutyric acid, butyric acid, isovaleric acid, and valeric acid in insomniac mice. Subsequent correlation analysis revealed that microbiota show obvious correlations with both biochemical markers and metabolites. ConclusionsThe results provide compelling evidence that ZSS-PR effectively mitigates abnormal activity, reduces cerebral pathological changes, and restores abnormal levels of neurotransmitters, endocrine hormones, and cytokines in mice with insomnia. The underlying mechanism is intricately linked to the modulation of gut microbiota and endogenous metabolic pathways.

Protective Effect of Egg Yolk Lipids against Dextran Sulfate Sodium-Induced Colitis: The Key Role of Gut Microbiota and Short-Chain Fatty Acids.

Egg yolk lipids significantly alleviate dextran sulfate sodium (DSS)-induced colitis by inhibiting NLRP3 inflammasome, reversing gut microbiota dysbiosis, and increasing short chain fatty acids (SCFAs) concentrations. However, the role of gut microbiota and the relationship between SCFAs and NLRP3 inflammasome are still unknown. Here, this study confirms that antibiotic treatment abolishes the protective effect of egg yolk lipids on DSS-induced colonic inflammation, intestinal barrier damage, and lipopolysaccharide translocation. Fecal microbiota transplantation also supports that egg yolk lipids alleviate colitis in a gut microbiota-dependent manner. Then, the study investigates the relationship between SCFAs and NLRP3 inflammasome, and finds that SCFAs significantly suppress colitis via inhibiting colonic NLRP3 inflammasome activation and proinflammatory cytokines secretions (interleukin, IL)-1β and IL-18, and combined treatment of SCFAs and MCC950 (NLRP3 inhibitor) shows a better activity against colitis and NLRP3 inflammasome activation. Together, these findings provide positive evidence for gut microbiorta-SCFAs-NLRP3 axis as a novel target involving in the therapy of inflammatory bowel disease.

Diet changes due to urbanization in South Africa are linked to microbiome and metabolome signatures of Westernization and colorectal cancer

Transition from traditional high-fiber to Western diets in urbanizing communities of Sub-Saharan Africa is associated with increased risk of non-communicable diseases (NCD), exemplified by colorectal cancer (CRC) risk. To investigate how urbanization gives rise to microbial patterns that may be amenable by dietary intervention, we analyzed diet intake, fecal 16 S bacteriome, virome, and metabolome in a cross-sectional study in healthy rural and urban Xhosa people (South Africa). Urban Xhosa individuals had higher intakes of energy (urban: 3,578 ± 455; rural: 2,185 ± 179 kcal/d), fat and animal protein. This was associated with lower fecal bacteriome diversity and a shift from genera favoring degradation of complex carbohydrates (e.g., Prevotella) to taxa previously shown to be associated with bile acid metabolism and CRC. Urban Xhosa individuals had higher fecal levels of deoxycholic acid, shown to be associated with higher CRC risk, but similar short-chain fatty acid concentrations compared with rural individuals. Fecal virome composition was associated with distinct gut bacterial communities across urbanization, characterized by different dominant host bacteria (urban: Bacteriodota; rural: unassigned taxa) and variable correlation with fecal metabolites and dietary nutrients. Food and skin microbiota samples showed compositional differences along the urbanization gradient. Rural-urban dietary transition in South Africa is linked to major changes in the gut microbiome and metabolome. Further studies are needed to prove cause and identify whether restoration of specific components of the traditional diet will arrest the accelerating rise in NCDs in Sub-Saharan Africa.

Lactic acid bacteria reduce bacterial diarrhea in rabbits via enhancing immune function and restoring intestinal microbiota homeostasis.

Numerous previous reports have demonstrated the efficacy of Lactic acid bacteria (LAB) in promoting growth and preventing disease in animals. In this study, Enterococcus faecium ZJUIDS-R1 and Ligilactobaciiius animalis ZJUIDS-R2 were isolated from the feces of healthy rabbits, and both strains showed good probiotic properties in vitro. Two strains (108CFU/ml/kg/day) were fed to weaned rabbits for 21 days, after which specific bacterial infection was induced to investigate the effects of the strains on bacterial diarrhea in the rabbits. Our data showed that Enterococcus faecium ZJUIDS-R1 and Ligilactobaciiius animalis ZJUIDS-R2 interventions reduced the incidence of diarrhea and systemic inflammatory response, alleviated intestinal damage and increased antibody levels in animals. In addition, Enterococcus faecium ZJUIDS-R1 restored the flora abundance of Ruminococcaceae1. Ligilactobaciiius animalis ZJUIDS-R2 up-regulated the flora abundance of Adlercreutzia and Candidatus Saccharimonas. Both down-regulated the flora abundance of Shuttleworthia and Barnesiella to restore intestinal flora balance, thereby increasing intestinal short-chain fatty acid content. These findings suggest that Enterococcus faecium ZJUIDS-R1 and Ligilactobaciiius animalis ZJUIDS-R2 were able to improve intestinal immunity, produce organic acids and regulate the balance of intestinal flora to enhance disease resistance and alleviate diarrhea-related diseases in weanling rabbits.

The Changes in Fecal Bacterial Communities in Goats Offered Rumen-Protected Fat.

Leizhou goats are famous for their delicious meat but have inferior growth performance. There is little information on rumen-protected fat (RPF) from the Leizhou goat. Hence, we observed the effects of RPF on growth, fecal short-chain fatty acids, and bacteria community with respect to Leizhou goats. Twelve goats (13.34 ± 0.024 kg) were selected and assigned randomly to one of two treatments: (1) a control diet (CON) and (2) 2.4% RPF with a control diet (RPF). The final body weight and average daily gain (ADG) were greater (p < 0.05), and the dry matter intake (DMI): ADG was lower (p < 0.05) in the RPF group than in the CON group. There were no differences in DMI between the CON and RPF groups. The concentrations of total short-chain fatty acids, acetate, propionate, and butyrate were lower (p < 0.05) in the RPF group than in the CON group. The relative abundances of Ruminococcus, Rikenellaceae_RC9_gut_group, Treponema, norank_f__norank_o__RF39, Eubacterium_siraeum_group, and Ruminococcus_torques_group were lower (p < 0.05) in the RPF group than in the CON group. The relative abundances of Bacteroides, norank_f__norank_o__Clostridia_UCG-014, norank_f__Eubacterium_coprostanoligenes_group, Eubacterium_ruminantium_group, norank_f__Oscillospirale-UCG-010, Oscillospiraceae_UCG-002, and Family_XIII_AD3011_group were greater (p < 0.05) in the RPF group than in the CON group. It was concluded that RPF could improve the goats' growth performance by regulating their fecal bacteria communities.

Green banana biomass anti-obesogenic, anti-hyperlipidemic, antidiabetic, and intestinal function potential effects: a systematic review.

Apparently, the consumption of resistant-starch food sources, such as green banana biomass, stimulates the proliferation of short-chain fatty acid intestinal bacteria producers, which can contribute to intestinal health and reduce the risk of chronic diseases. However, the available scientific evidence is scarce and no study has systematically evaluated such evidence. The aim of this study was to analyze the potential effects of green banana biomass on anthropometry, body composition, and biochemical and intestinal variables in humans and animals. The Cochrane Library, Embase, Medline/PubMed, Scopus, and Web of Science electronic databases were searched in January 2024 for eligible articles. Studies that tested the effects of cooked peeled or unpeeled green banana on anthropometric, biochemical, and/or intestinal variables were included. This systematic review was conducted according to the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines. The classification and assessment of the quality of studies were based on the relevant criteria related to the design of these studies and the quality criteria checklist of the Academy of Nutrition and Dietetics manual. Twelve studies published between 2001 and 2021 were included in the review. The results of human studies indicate that the ingestion of green banana biomass controlled intestinal dysfunction (50-300 g/day for 5-14 days or 30 g/day for 8 wk) in children, and showed potential anti-obesogenic, anti-hyperlipidemic, and antidiabetic (40 g/day for 24 wk) effects in adults. In rats, biomass consumption led to potential anti-obesogenic (25 g/day for 8 wk), anti-hyperlipidemic, and antidiabetic (∼8-30 g/day for 12 wk) effects. Consumption of green banana biomass seems to exert beneficial effects on intestinal function and potential effects on obesity, dyslipidemia, and diabetes. These effects may be related to increased fecal short-chain fatty acid concentrations as a result of type 3 resistant starch present in biomass. Ingredients in Rhus chinensis Mill. fruits oil relieve fatigue by reducing oxidative damage and regulating energy metabolism and gut microbiota

Fatigue is an intricate physiological phenomenon, causing serious secondary concerns and bioregulation disorders. The objective of this research was to look into the anti-fatigue effect and potential mechanisms of natural ingredients in methanol extract of edible oil from Rhus chinensis Mill. fruits (PMRO). Results showed that a total of 18 fatty acids and 9 anacardic acids were identified by GC-MS and UHPLC-ESI-HCES/MS in PMRO. PMRO significantly increased the weight-bearing swimming time. Further investigations showed that PMRO significantly improved physiological indexes related to fatigue and regulated the oxidative stress (Keap1/Nrf2) and energy metabolism (AMPK/PGC-1α)-related proteins expression to exert anti-fatigue effects. The 16S rRNA sequence analysis indicated that a potential role of PMRO in modulating the abundance of gut bacteria and influencing the short-chain fatty acids content, thereby exerting an anti-fatigue effect. Therefore, the natural ingredients in PMRO may hold promise as a dietary supplement capable of delaying body fatigue.

Efficacy of high-intensity interval and continuous endurance trainings on cecal microbiota metabolites and inflammatory factors in diabetic rats induced by high-fat diet.

Physical exercise is known to modulate the intestinal microbiota composition and control the symptoms of metabolic syndrome. In this research, we intend to investigate and compare the effect of high-intensity interval and continuous endurance trainings (HIIT and CET) on cecal microbiota metabolites and inflammatory factors in diabetic rats. A number of Wistar rats were made diabetic by a high-fat diet and trained under two types of exercise protocols, HIIT and CET. After taking samples from the cecal tissue and serum of rats to reveal the effect of exercise, three microbial species from the Firmicute and Bacteroid phyla, which are the main types of intestinal microbes, and their metabolites include two short-chain fatty acids (SCFAs), butyrate and propionate and also, the inflammatory factors TLR4 and IL6 were analyzed through quantitative polymerase chain reaction (qPCR), high-performance liquid chromatography (HPLC), and Enzyme-linked immunosorbent assay (ELISA) methods. In general, exercise while increasing the representative of Firmicute has caused a relative reduction of Bacteroides and improved the concentration of SCFAs. In this regard, HIIT outperforms CET in up-regulating Akkermansia and Butyrivibrio expression, and butyrate and propionate metabolites concentration. Also, both exercises significantly reduced cecal expression of TLR4 and sera concentration of IL6 compared to the diabetic group, although the reduction rate was higher in the CET group than in HIIT. Our findings suggest that some symptoms of metabolic syndrome such as intestinal dysbiosis and the resulting metabolic disorders are better controlled by HIIT and inflammation by CET. Certainly, more extensive research on other contributing factors could help clarify the results.
